Highlights • A novel STEC LAMP assay was developed for the detection of stx 1 and stx 2. • Three detection strategies were evaluated, real-time fluorescence acquisition and two naked-eye colorimetric ones. • Four simple DNA extraction and purification protocols suitable for point-of-care testing were compared. • The “glass milk” protocol demonstrated to be the best option considering complexity, cost, turnaround time and performance. • An LOD50 <10 CFU/ 25 g, with high diagnostic sensitivity, specificity and accuracy in spiked RTE salad samples was reached.
